Long-term preservation protocol for filamentous fungi

A conidial suspension of sporulating filamentous fungi is prepared in an appropriate cryoprotectant solution such as DMSO or glycerol / skim milk. A stepwise freezing rate to minimize intracellular ice crystal formation is used. The samples are cooled very rapidly from a given temperature and stored in the gas phase of liquid nitrogen.

Fungal cultures that do not sporulate can also be stored in the gas phase of liquid nitrogen. Mycelia containing plugs cut from agar cultures are placed in vials containing glycerol and stored at minus 196°C.
